Better treatment for brain cancer revealed by new molecular insights

Nearly a third of adults with the most common type of brain cancer develop recurrent, invasive tumors after being treated with a drug called bevacizumab. The molecular underpinnings behind these detrimental effects have now been published by Cell Press in the July issue of Cancer Cell. The findings reveal a new treatment strategy that could reduce tumor invasiveness and improve survival in these drug-resistant patients.

"Understanding how and why these tumors adopt this invasive behavior is critical to being able to prevent this recurrence pattern and maximizing the benefits of bevacizumab," says study author Kan Lu of the University of California, San Francisco (UCSF).

Glioblastoma multiforme (GBM) is the most aggressive type of tumor originating in the brain. GBM tumors express high levels of vascular endothelial growth factor (VEGF), a protein that promotes the growth of new blood vessels that provide nutrients that allow tumors to expand. In 2009, the Food and Drug Administration approved bevacizumab, a VEGF inhibitor, for GBM patients who don't respond to first-line therapies. Although the drug is initially effective, up to 30% of patients develop tumors that infiltrate deep into the brain, making surgery and treatment difficult.

To study how bevacizumab can lead to adverse effects, senior study author Gabriele Bergers of UCSF and her collaborators focused on hepatocyte growth factor (HGF), a protein that controls the growth and movement of cells, because they previously found a link between VEGF and HGF in GBM cells. In the new study, they found that VEGF inhibits the migration of GBM cells by decreasing HGF signaling through its receptor MET. Moreover, tumors were much less invasive?and survival improved?in mice with GBM tumors lacking both VEGF and MET rather than just VEGF alone. The results suggest that MET plays a critical role in GBM invasion when VEGF is blocked.

"These findings provide a rationale for therapeutically combining VEGF and MET inhibition so that patients can benefit from bevacizumab without developing more invasive tumors," Lu says. Because the VEGF and HGF/MET signaling pathways are active in a variety of tumors, this combined treatment strategy may also be applied to other types of cancer.

More charges in murder tied to ?Fast and Furious?

The Justice Department Monday unveiled new charges against Ivan Soto-Barraza and four others for the murder of Border Patrol Agent Brian Terry.

Federal prosecutors on Monday revealed more charges in the murder of U.S. Border Patrol Agent Brian Terry.

The indictment, unsealed Monday in Tuscon, Ariz., charges five men with involvement in Terry's death, which is at the center of a controversy over a Bureau of Alcohol, Tobacco and Firearms and Explosives (ATF) gun tracking operation known as "Fast and Furious."


According to the indictment, Manuel Osorio-Arellanes, Jesus Rosario Favela-Astorga, Ivan Soto-Barraza, Heraclio Osorio-Arellanes and Lionel Portillo-Meza are charged with crimes including first degree murder, second degree murder, conspiracy to interfere with commerce by robbery, attempted interference with commerce by robbery, use and carrying a firearm during a crime of violence, assault on a federal officer and possession of a firearm by a prohibited person.??

The 11-count indictment alleges that on Dec. 14, 2010, five of the defendants engaged in a firefight with Border Patrol agents. Terry was shot and killed in the gunfire.

The defendants illegally entered the United States from Mexico to rob drug traffickers of their contraband, according to the indictment.

In addition to the murder of Terry, the indictment also alleges that the five defendants assaulted Border Patrol Agents William Castano, Gabriel Fragoza and Timothy Keller, who were with Terry during the shootout.

A sixth defendant, Rito Osorio-Arellanes, is charged only with conspiracy to interfere with commerce by robbery.

Two of the individuals involved with the shooting are already in custody: Manuel Osorio-Arellanes was arrested on the night of the shooting and Rito Osorio-Arellanes was arrested by Border Patrol agents on December 12, 2010, on immigration charges.

The FBI is offering a reward of up to $1 million for information leading to the arrest of the remaining fugitives.

"We will stop at nothing to bring those responsible for his murder to justice," Attorney General Eric Holder said in a statement.?"Today's announcement reflects the department's unrelenting commitment to finding and arresting the other individuals responsible for this horrific tragedy so that Agent Terry's family, friends and fellow law enforcement agents receive the justice they deserve."

MIPS unveils new Aptiv mobile chip design, vows more speed in a tiny space

MIPS has only had a slight amount of traction in smartphones and tablets given that its best friend is Ainovo. It might get a better footing in an ARM-dominated world with a new range of processors in the Aptiv line. The highest-end chip of the bunch, the ProAptiv, is reportedly much smaller than its ARM opponents across the aisle while managing the same speed, goosing the floating-point math, memory addressing and multi-core support to where it's as much as 75 percent faster over the previous generation. That's good news for mobile devices and set-top boxes where space is tight. MIPS isn't leaving embedded gear like 4G LTE modems alone, either, as the mid-size InterAptiv and tiny MicroAptiv are getting similar improvements. If you're eager to get cracking on a processor of your own, you can license MIPS' new work right away -- and us plebeians who simply buy the devices can expect Aptiv to be ready for gadget use by the middle of the year.

Facebook has been making a lot of promises during a tour to drum up interest in its ever-nearing IPO, but the one gadget-heads have been wanting to hear the most, a commitment to its mobile apps, has been elusive -- until now. Everyone's favorite hooded CEO, Mark Zuckerberg, is telling investors in his home 'burg of the San Francisco Bay that mobile is front and center in his company's plans. We're hoping that means new app features, although Zuck is likely referring to money-making as well: shareholders are jittery knowing that Facebook makes most of its money on web ads that it's not running on smartphones and tablets. Paid titles in App Center will go a long way towards scratching that itch, mind you. As for us, we'll just be happy if Facebook takes less than a year and a half to produce a major tablet app.
